Colocalization of the AhR and ARNT within the nucleus was seen in the lower epidermal area. In the control group, the AhR Alizarin and ARNT were weakly stained in the epidermis and there was no colocalization of the two molecules (Fig. S1). The expression levels of the AhR, ARNT and CYP1A1 mRNA were higher in the AD group than in the control group (Fig. S2a). When NHEK was treated with TCDD, the AhR showed high signal intensity in the nucleus at three and 6 h after the treatment and did the ARNT. In conclusion, the results of this study suggest that the AhR is highly expressed in the acute lesional skin of patients with AD and psoriasis and that activation of the AhR increases the expression of inflammatory cytokines in NHEK. As this is only a pilot study, we are planning on further studies with increased sample size, and additional studies such as immunoblot analysis, to investigate the role of AhR.
