Towards the posterior end of the VNC, Capa expression declined (A7), and in A8 no Capa could be found, though the presence of the Va neurons could be confirmed via Dac/Dimm. This, together with the fact that Va neurons in A1, the most anterior abdominal segment, never express Capa ( Fig.?2A) indicates that factors responsible for the differentiation towards the peptidergic fate of Va neurons are missing in these segments of the VNC. Hence, in addition to PCD, the segmental patterning of Va neurons is guided by other cues. The thoracic segments T1 to T3 of late embryos, even in H99 homozygotes, did not show any Va cells positive for either Capa or Dac/Dimm, which parallels the diglyceride situation in wild type (Fig. 2ACB). This, together with the lack of Caspase-3 staining, GDC-0941 datasheet favors the idea that PCD is not responsible for the disappearance of thoracic Va neurons. In order to investigate which of the death activator genes are responsible for the apoptosis of the abdominal Va neurons, smaller deletions were analyzed (Fig.?2). In X14 hid is missing, X25 deletes hid and grim and XR38 deletes the reaper gene ( Peterson et al., 2002). The single deletions of either hid or reaper showed a small, but statistically significant effect ( Fig.?2F; P?=?0.01 and P?AZD0530 order (data not shown). However, the possibility that hid and grim act synergistically cannot be excluded since both genes are removed by the X25 deficiency. Hox genes are known to specify the identity of body segments along the anteriorCposterior axis in most animals (Hirth et al., 2001, Hirth and Reichert, 1999, Rogulja-Ortmann and Technau, 2008?and?Sprecher et al., 2004), and several recent studies have demonstrated their involvement in segment-specific neurogenesis (Bello et al., 2003, Berger et al., 2005b?and?Karlsson et al., 2010). In the Drosophila VNC, the Hox genes are represented by Antennapedia (Antp), Ultrabithorax (Ubx), abdominal A (abd-A) and Abdominal-B (Abd-B), which are expressed in overlapping anteriorCposterior domains ( Hirth et al., 1998).
