We applied Scanning Natural Product Library Electron Microscope to evaluate the morphological features of endothelial cells on titanium substrates. The mRNA expression of NGF and its receptors (TrkA, p75) and angiogenic related genes (VEGF) were measured by real-time PCR. For in vivo assay, each groups�� cell culture supernatants were added to Chicken embryo chorioallantoic membrane (CAM) of 8th day fertilized eggs and cultured for another 24?h. Then the CAMs were fixed and transfer each specimen to petri dishes. The images were acquired and analyzed with Image Pro Plus. Results NGF could induce ECs proliferation on titanium substrates from day 3 to 7. For the morphological features, ECs are ball-like on titanium surface at 2?h. While at 24?h, the cells were totally stretched and fused into the rough titanium backgrounds. Nerve growth factor could up regulate the mRNA expression levels of NGF, TrkA, p75, VEGF. In vivo, the supernatants of NGF group could induce more neovascularization in CAM membrane. Conclusion and clinical implications This study first time demonstrated that NGF could enhance ECs proliferation, genes expression on titanium surface and more neovascularization in CAM.
