Functionally, rats with CHF showed significantly increased left ventricular end-diastolic pressure compared with their sham-operated counterparts (Table 1). To compare the expression of transcripts encoding the CX3CR1 receptor in sham-operated rats and rats with CHF, we performed quantitative RT-PCR analysis on mRNA extracted from the PVN. We found that the transcript for CX3CR1 was significantly increased in the PVN of rats with CHF compared with sham-operated rats (n= 5 per group; Fig. 3A). Western blot analysis showed that CX3CR1 protein expression is significantly upregulated in the PVN (Fig. 3B) but not cortex (Fig. 3C) of rats with heart failure compared with their control, Ficain sham-operated counterparts. In order to study the cardiovascular effects of FKN within the PVN, we performed unilateral microinjections of increasing doses of FKN in anaesthetized rats. Unilateral microinjections of FKN into the PVN (1.0, 2.5, 5.0 and 7.5 nmol in 50 nl) elicited a dose-related decrease in MAP, ranging from ?5 �� 2 (1.0 nmol FKN) to ?32 �� 4 mmHg (7.5 nmol FKN), with a dose-related increase in HR ranging from 7 �� 3 to 27 �� 5 beats min?1 (Fig. 4). Based on this dose�Cresponse curve, a dose of 2.5 nmol was chosen as the optimal dose, because higher doses tended to produce irreversible responses whereby basal blood pressure could not be re-established.
